Steigern Sie Durchsatz und Robustheit Ihrer Antikörperaufreinigung TOYOPEARL AF-rProtein A-650F vereint eine hohe Proteinbindekapazität bei hohen Flussraten mit einer guten Stabilität gegenüber alkalischen cleaning-in-place (CIP) Reinigungslösungen. Protein A Affinitätschromatographie ist die Standardmethode für das sogenannte Capturing von monoklonalen Antikörpern (mAb) bei der Aufreinigung aus Zellkulturüberständen. In den letzten Jahren konnten die Fermentationstiter der Antikörper stetig gesteigert werden, so dass inzwischen die Kapazitätsengpässe vor allem bei der Aufreinigung der Zielmoleküle auftreten. TOYOPEARL AF-rProtein A-650F wurde mit dem Ziel entwickelt, diesen Engpass zu beheben. Es besitzt auch bei einer kurzen Kontaktzeit eine hohe dynamische Bindekapazität und ermöglicht so eine schnellere Beladung der Säule und eine entsprechende Steigerung des Gesamtdurchsatzes. Für dieses Affinitätsmedium wurde ein neuentwickelter, rekombinanter Ligand an den bekannte Methacrylat Basispartikel der TOYOPEARL Medien gebunden. Die Alkalistabilität des Liganden wurde durch gezielte Mutationen erhöht. Er ist durch mehrere Bindungen mit der Matrix verbunden und bietet dadurch eine höhere chemische und thermische Stabilität. In der Praxis wirkt sich das in einem geringeren Protein A Leaching und einer hohen Stabilität gegenüber basischen Waschlösungen aus, die typischerweise in CIP- Prozeduren eingesetzt werden. Aufgrund seiner relativ starren Polymermatrix besitzt das Material außerdem ausgezeichnete Druck/Flusseigenschaften und ist daher nicht nur im Labormaßstab sondern auch besonders gut im industriellen Produktionsmaßstab einsetzbar. Toyopearl GigaCap S-650

Toyopearl GigaCap S-650 is a high capacity, high resolution cation exchange resin* optimized for process scale applications, including the capture and purification of IgG as well as the polishing of monoclonal antibodies. A Toyopearl HW-65 polymeric bead has been chemically modified to provide a higher number of cationic binding sites for increased resin capacity. While Toyopearl GigaCap S-650M was specifically optimized for IgG, with dynamic binding capacities in the vicinity of 140 mg/mL for IgG, it also exhibits high binding capacities for smaller proteins. Typical dynamic binding capacities for lysozyme are in excess of 160 mg/mL. Toyopearl GigaCap S-650S has dynamic binding capacities approaching 165 g/L for gamma globulin. This high capacity, combined with the 35 µm mean particle size, makes Toyopearl GigaCap S-650S ideal for high resolution applications. Toyopearl GigaCap S-650 media is stable at high linear velocities, has good pressure-flow characteristics and will withstand back pressures up to 3 bar. The higher capacity and back pressure stability of Toyopearl GigaCap S-650 resin creates opportunities for increased throughput in downstream cation exchange purification steps. Automated LIII Lipoprotein Analyzer HLC-729LP II

Tosoh Begins Sales of Automated LIII Lipoprotein Analyzer HLC-729LP II Tosoh has announced the January start of sales of its new lipoprotein analyzer, the HLC-729LP II. This automated LIII lipoprotein analyzer boasts speedy and accurate measurements of high-density lipoprotein (HDL), of low-density lipoprotein (LDL), and of very low-density lipoprotein (VLDL) cholesterol—important clinical tests for the diagnosis and treatment of high cholesterol. The HLC-729LP II demonstrates superior separation capabilities and analytical precision compared with the popular electrophoresis method. Tosoh’s HLC-729LP II lipoprotein analyzer measures lipoproteins in serum using a high-performance liquid chromatography (HPLC) process. That process separates the different densities of cholesterol and measures their concentration based on coloring elicited using an enzymatic reaction. Individual tests take only 5.2 minutes and provide accurate typing of high blood cholesterol based on the World Health Organization (WHO)’s classifications. The diagnostic reagent used in testing is the HLC-Test Tosoh Lipopack. It is estimated that up to 22 million Japanese have high blood cholesterol, putting that condition on a par with diabetes as a major lifestyle disease. HDL cholesterol is considered “good” cholesterol because it prevents hardening of the arteries, while LDL cholesterol is known as “bad” cholesterol because it causes hardening of the arteries. Recently, VLDL cholesterol levels in the blood have also drawn attention because of the link between high VLDL levels and the risk of hardening of the arteries. VLDL levels increase with obesity, metabolic syndrome, and insulin resistance. The advent of Tosoh’s HLC-729LP II lipoprotein analyzer adds greatly to the ability to test for what is a growing problem worldwide.
